The IGB-MarabanX Plasmid Extraction Kit utilizes a modified alkaline lysis method combined with advanced silica-based spin-column technology.
The process involves three key steps:
  • Cell Lysis: Bacterial cells are lysed using an optimized alkaline lysis buffer system, which selectively denatures chromosomal DNA while keeping plasmid DNA intact. RNase A is included to remove RNA contaminants, ensuring high-purity DNA.
  • DNA Binding:   The lysate is loaded onto a silica-membrane spin column, where plasmid DNA selectively binds in the presence of chaotropic salts, while impurities like proteins and genomic DNA are washed away.
  • Elution:    Pure plasmid DNA is eluted in a low-salt buffer or water (pH 7.0–8.5), ready for immediate use in downstream applications.

    Before use, add the provided RNase A to Buffer P1 and store at 4°C for up to 3 months. Add ethanol to Buffer PW to achieve an 80% v/v concentration, as recommended in protocols.

    For low-copy plasmids, yields can be enhanced by adding chloramphenicol (170 mg/L) to the culture medium to amplify copy numbers.
